Production of Alkaline Protease Using Industrial Waste Effluent as Low-cost Fermentation Substrate

Alkaline proteases are the most important groups of commercial enzymes, which have been broadly used in industrial processes. In this study, Bacillus sp. PTCC 1538 was selected as a biological agent to produce alkaline protease. Enzyme production under submerge fermentation using waste effluent investigated. Since costs raw material plays an role cost enzyme production, corn steep liquor (CSL) low-cost substrate reduce production. Various carbon sources were auxiliary substrates enhance Results showed that maximum activity obtained when wheat bran substrate. Optimal media composition and growth conditions for protease defined. The optimum found be pH 8, incubation temperature 37 °C, CSL inoculum size 5 v/v %, yeast extract concentrations 2 6 g/l, respectively. CaCl2 activator proteolytic enzyme. Under condition, 100.7 U/ml at concentration 1.5 g/l.
